A practical reference on intranasal delivery: what it is, how it behaves, what the literature reports, and where the honest uncertainties sit.

Peptide bonds are vulnerable to protease attack, and Selank is no exception. Measured half-life in serum is short, on the order of minutes in several reports, which explains why intranasal administration is the common route described in the literature. Absorption across the nasal mucosa partially bypasses first-pass hepatic metabolism. Quantitative data on human bioavailability remain limited and are difficult to compare across studies.
Lyophilised material kept dry at minus 20 degrees Celsius or colder is the most stable form, and suppliers commonly state a shelf life of two years or more under those conditions. Once dissolved, degradation accelerates through hydrolysis and deamidation, particularly at alkaline pH or elevated temperature. Working solutions are usually divided into single-use aliquots to avoid repeated freeze-thaw cycles. The choice of reconstitution solvent affects both stability and the ionic strength of the final preparation.
Selank is a synthetic heptapeptide with the sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro, frequently abbreviated as TKPRPGP. It was designed as a structural analogue of tuftsin, a naturally occurring tetrapeptide released by enzymatic cleavage of the immunoglobulin heavy chain. The two additional proline residues at the C-terminal end extend the parent chain and change how the molecule behaves in solution. The free peptide has a calculated molecular mass of approximately 751.9 g/mol and is generally supplied as a lyophilised white to off-white powder.
Development work on the compound began in the 1980s and 1990s at the Institute of Molecular Genetics in Moscow, within the same research programme that produced the peptide Semax. Early investigators sought a tuftsin derivative with improved resistance to enzymatic breakdown and with activity in the central nervous system after peripheral administration. Most of the primary literature from this period was published in Russian, a factor that still shapes how easily the findings can be checked by outside groups.
| Property | Value | Notes |
|---|---|---|
| Dry storage temperature | -20 C or below | Sealed, desiccated, protected from light |
| Solution stability | Hours to days at 2-8 C | Depends on buffer and microbial load |
| Preferred pH range | Near neutral to slightly acidic | Alkaline pH accelerates hydrolysis |
| Main degradation routes | Hydrolysis and deamidation | No cysteine or methionine in sequence |
| Identity confirmation | Electrospray mass spectrometry | Observed mass compared with 751.9 Da |
Purity assessment relies mainly on reverse-phase high-performance liquid chromatography with ultraviolet detection. Because the peptide lacks a strong chromophore, detection often uses backbone absorbance near 214 nm. Identity is confirmed by mass spectrometry, typically electrospray ionization or matrix-assisted laser desorption, comparing the measured mass against the expected value. Amino acid analysis can verify composition after acid hydrolysis. Diastereomer content and residual counterions are reported less often, although both can influence biological assays.
Lyophilized material is generally stable for extended periods when kept dry at or below minus twenty degrees Celsius. Working solutions are less stable, and common practice is to aliquot and freeze them so that repeated freeze-thaw cycles are avoided. Aqueous solutions are sensitive to pH extremes and to microbial growth, so short-term storage at refrigerator temperature is typical. Oxidation and hydrolysis are the principal degradation routes. Reconstitution with sterile water or a mild buffer is standard, and solutions should be protected from light.
The primary structure of Selank is Thr-Lys-Pro-Arg-Pro-Gly-Pro, corresponding to the molecular formula C33H57N11O9 and a monoisotopic mass of roughly 751.9 daltons. The N-terminal threonine and the arginine residue in the fourth position are shared with tuftsin, which carries the sequence Thr-Lys-Pro-Arg. The three additional residues at the C-terminus, Pro-Gly-Pro, extend the chain and are associated with greater resistance to enzymatic degradation. This extension also separates Selank from the shorter parent peptide.
Naming conventions place Selank in the same research family as Semax, another Russian-developed peptide investigated for cognitive effects. The two compounds share a lineage but differ in sequence and in the biological systems proposed as their targets. Semax descends from ACTH fragments, whereas Selank descends from tuftsin. Publications sometimes identify Selank by its full peptide sequence or by laboratory codes rather than one uniform trade name. Because replication outside Russia is limited, reports on its properties are best read alongside the study design and the purity of the material tested.
=== Abiogenic petroleum === An alternative mechanism to the one described above was proposed by Russian scientists in the mid-1850s, the hypothesis of abiogenic petroleum origin (petroleum formed by inorganic means), but this is contradicted by geological and geochemical evidence. Abiogenic sources of oil have been found but never in commercially profitable amounts. "The controversy isn't over whether abiogenic oil reserves exist," said Larry Nation of the American Association of Petroleum Geologists. "The controversy is over how much they contribute to Earth's overall reserves and how much time and effort geologists should devote to seeking them out."
=== Volatile content determination in geological materials === FTIR spectroscopy is often used in geology to quantify volatile species, H2O and CO2, in minerals, glasses, and melt inclusions. Quantifying the concentrations of these volatiles is important, as these volatiles influence magma storage conditions, crystallization, degassing, and eruption style — all of which modulate the properties of magmas such as density and viscosity. In silicate glasses and melt inclusions, absorbance bands in the near- and mid-IR associated with H2O (dissolved in melts as OH- or H2O) and CO2 (dissolved in melts as CO32- or CO2) can be quantified into concentrations with the Beer-Lambert Law. In nominally anhydrous minerals, trace amounts of H+ are measured to understand mantle water storage and other magmatic processes.
Cholecystokinin tetrapeptide (CCK-4, tetragastrin, Trp-Met-Asp-Phe-NH2) is a peptide fragment derived from the larger peptide hormone cholecystokinin. Unlike cholecystokin which has a variety of roles in the gastrointestinal system as well as central nervous system effects, CCK-4 acts primarily in the brain as an anxiogenic, although it does retain some GI effects, but not as much as CCK-8 or the full length polypeptide CCK-58. CCK-4 reliably causes severe anxiety symptoms when administered to humans in a dose of as little as 50 μg, and is commonly used in scientific research to induce panic attacks for the purpose of testing new anxiolytic drugs. Since it is a peptide, CCK-4 must be administered by injection, and is rapidly broken down once inside the body so has only a short duration of action, although numerous synthetic analogues with modified properties are known. Albiglutide (trade names Eperzan in Europe and Tanzeum in the US) is a glucagon-like peptide-1 agonist (GLP-1 agonist) drug marketed by GlaxoSmithKline (GSK) for treatment of type 2 diabetes. In 2017 GSK announced Albiglutide's withdrawal from the worldwide market for economic reasons, and remaining stocks in the supply chain were effectively depleted by 2018.
The non-essential amino acid arginine, consumed in sufficient amounts, is thought to act as a donor for the synthesis of nitric oxide, a vasodilator. A review confirmed blood pressure lowering. Taurine, a popular dietary supplement ingredient with claims made for sports performance, is technically not an amino acid. It is synthesized in the body from the amino acid cysteine.

=== Precambrian bodies === The Earth formed around 4.6 billion years ago, with unicellular life emerging somewhat later after the cessation of the Late Heavy Bombardment, a period of intense asteroid impacts possibly caused by migration of the gas giant planets to their current orbits, however multicellularity and bodies are a relatively recent event in Earth's history. Bodies first started appearing towards the end of the Precambrian Era, during the Ediacaran period. The fossils of the Ediacaran period were first found in Southern Australia in the Ediacara Hills, hence the name. However these fossils were initially thought to be part of the Cambrian and it wasn't until the late 1950s when Martin Glaessner identified the fossils as actually being from the Precambrian era. The fossils that were found date to about 600 million years ago and are found in a variety of morphologies.
The quadriplegia dissipates once the patient lies supine. An extremely rare complication of sCSFL is third nerve palsy, where the ability to move one's eyes becomes difficult and interrupted due to compression of the third cranial nerve. There are documented cases of reversible frontotemporal dementia and coma. Coma due to a CSF leak has been successfully treated by using blood patches and/or fibrin glue and placing the person in the Trendelenburg position. Empty sella syndrome, a bony structure that surround the pituitary gland, occurs in CSF leak patients.
A transdermal gel was formulated to address this but plasma concentrations were undetectable. In cats oral tramadol at 6 mg/kg provides equivalent analgesia to intramuscular tramadol at 4 mg/kg. One study comparing tramadol to meperidine found tramadol to be more effective at providing analgesia for cats undergoing ovariohysterectomy. A study looking at treatment of osteoarthritis in geriatric cats found owner-reported improvements but also significant adverse effects. Reported adverse effects of tramadol include: diarrhoea, dysphoria, anorexia, tachycardia, pyrexia, tachypnoea, and increased levels of serotonin leading to agitation. Tramadol has the potential to cause serotonin syndrome when administered alongside serotonin re-uptake inhibitors, monoamine oxidase inhibitors, and tricyclic antidepressants. In horses, tramadol is ineffective by itself. Two studies looking at intravenous tramadol found it provided no thermal or mechanical antinociception, another study looking at oral tramadol for chronic laminitis found it provided minimal analgesia, and another study looking at intraarticular tramadol found it provided no post-operative analgesia following arthroscopy; however, when administered with paracetamol (acetaminophen) tramadol provides short-acting mechanical antinociception and when administered with ketamine oral tramadol provides adequate analgesia for chronic laminitis. Based on this evidence tramadol is not recommended to be provided on its own for analgesia in horses.

Dry powder is best kept sealed, protected from light, and held at minus 20 degrees Celsius or below. Desiccant packaging helps limit moisture uptake because the material is hygroscopic. A sealed vial should be allowed to equilibrate to room temperature before opening to reduce condensation.
Hydrolysis of peptide bonds and deamidation of arginine and proline residues are the main degradation routes. Alkaline pH and higher temperatures accelerate both processes. Buffered, near-neutral solutions stored cold generally degrade more slowly than unbuffered ones.
Reverse-phase chromatography gives the clearest single view of related impurities and truncated sequences. Mass spectrometry then confirms that the main peak carries the expected mass. Neither test detects enantiomeric impurities, which require a separate chiral assay.
The sequence is Thr-Lys-Pro-Arg-Pro-Gly-Pro, commonly written as TKPRPGP. It shares the first four residues with tuftsin and carries three prolines in the chain. The proline-rich tail is the main structural feature that separates it from the parent tetrapeptide.
